ABSTRACT

Pancreatic adenocarcinoma is important in oncology because of its high mortality rate. Deaths may be avoided if an early diagnosis could be achieved. Several types of tumors overexpress gastrin-releasing peptide receptors (GRPr), including pancreatic cancer cells. Thus, a radiolabeled peptide derivative of gastrin-releasing peptide (GRP) may be useful as a specific imaging probe. The purpose of the present study was to evaluate the feasibility of using99mTc-HYNIC-βAla-Bombesin(7-14)as an imaging probe for Capan-1 pancreatic adenocarcinoma. Xenographic pancreatic tumor was developed in nude mice and characterized by histopathological analysis. Biodistribution studies and scintigraphic images were carried out in tumor-bearing nude mice. The two methods showed higher uptake by pancreatic tumor when compared to muscle (used as control), and the tumor-to-muscle ratio indicated that99mTc-HYNIC-βAla-Bombesin(7-14)uptake was four-fold higher in tumor cells than in other tissues. Scintigraphic images also showed a clear signal at the tumor site. The present data indicate that99mTc-HYNIC-βAla-Bombesin(7-14)may be useful for the detection of pancreatic adenocarcinoma.

ABSTRACT

Avaliou-se o efeito da inclusão de zeólita na dieta de bovinos de corte sobre os parâmetros ruminais, as digestibilidades total e parcial, a produção de nitrogênio, a eficiência microbiana e as características do sangue. Os tratamentos, com base na matéria seca (MS) da dieta, foram: 0; 0,75; 1,5; 2,25 e 3,0% de zeólita. Utilizaram-se cinco bovinos machos mestiços, fistulados no rúmen e abomaso, alimentados com silagem de milho e concentrado, representando 65% de volumoso e 35% de concentrado. O delineamento utilizado foi em quadrado latino 5×5, com cinco períodos de 15 dias. Os animais receberam 15g de dióxido de titânio (TiO2) do terceiro ao 12º dia de cada período. Houve efeito (P<0,05) sobre o consumo diário (kg/dia) de proteína bruta (PB) e efeito (P<0,05) sobre o consumo de MS e de fibra em detergente neutro corrigida para cinza e proteína, expresso em g/kg de peso. A digestibilidade total dos nutrientes digestíveis totais (NDT) foi influenciada pelos tratamentos (P<0,05). A digestibilidade ruminal dos carboidratos não fibrosos (CNF) foi afetada quadraticamente (P<0,05), assim como a intestinal da PB e dos CNF. A ingestão e a excreção fecal de nitrogênio (g/dia) foram influenciadas linearmente (P<0,05) pela inclusão de zeólita. Concluiu-se que a adição de zeólita na dieta, embora tenha aumentado os consumos de MS e de FDN, não melhorou a utilização da ureia em dietas de bovinos de corte.


We evaluated the effect of the inclusion of zeolite in the diet of beef cattle on the ruminal digestibility, total and partial production of nitrogen, microbial efficiency and blood characteristics. Treatments based on the dietary dry matter (DM) were: 0%, 0.75%, 1.5%, 2.25% and 3.0% of zeolite. We used five crossbred steers fistulated in the rumen and abomasum, fed corn silage and concentrate, representing 65% forage and 35% concentrate. The design was a 5x5 latin square with five periods of 15 days. The animals received 15g of titanium dioxide (TiO2) from the third to the 12th day of each period. A significant effect (P<0.05) on the daily consumption (kg/day) of crude protein (CP) was observed (P<0.05) for DM intake and neutral detergent fiber corrected for ash and protein (NDFap) expressed in g/kg. The total digestibility of total digestible nutrients (TDN) was influenced by treatments (P<0.05). Ruminal digestibility of non-fiber carbohydrates (NFC) was affected quadratically (P<0.05) as well as intestinal CP and NFC. Intake and fecal excretion of nitrogen (g/day) were affected linearly (P<0.05) by adding zeolite. It was concluded that the addition of zeolite in the diet, while increasing the intake of DM and NDF, did not improve the use of urea in the diet of beef cattle.

ABSTRACT

O trabalho teve como objetivo avaliar a influência de doses de cinzas em diferentes substratos no enraizamento de estacas de manjericão (Ocimum basilicum). O experimento foi conduzido em área do Núcleo de Capacitação e Pesquisa da Universidade Federal Rural da Amazônia, em Belém. Foi utilizado o delineamento experimental inteiramente casualizado, em esquema fatorial 2 x 5, constituído pelas combinações entre os fatores substratos (casca de arroz carbonizada e casca de castanha triturada) e acréscimo de cinzas (5 mL e 10 mL de cinza de casca de arroz, 5 mL e 10 mL de cinza de casca de castanha e sem cinza) com 4 repetições. Cada unidade experimental foi constituída por 10 estacas, num total de 400 estacas. Foram avaliadas as características percentual de enraizamento de estacas, número e comprimento de raízes. Não houve interações significativas entre os fatores avaliados (substrato e cinzas). Entre as características avaliadas o substrato casca de arroz carbonizada apresentou diferenças apenas no comprimento da raiz e no número de raízes, enquanto as dosagens de cinzas apresentaram diferenças apenas no percentual de enraizamento. O trabalho permitiu concluir que as dosagens de cinzas apresentaram efeito positivo sobre o enraizamento das estacas de manjericão, assim como o substrato a base de casca de arroz carbonizada propiciou melhores condições para o desenvolvimento das raízes. Assim, a casca de arroz é uma alternativa totalmente viável, pois além de apresentar características apropriadas para o enraizamento também possui baixo custo de aquisição, estando ao alcance de muitos produtores.


The study aimed to evaluate the influence of ash doses on different substrates on the stem rooting of basil (Ocimum basilicum). The experiment was conducted in the area of core training and research at the Universidade Federal Rural da Amazônia, in Belém, used a completely randomized design, factorial scheme 2 x 5 consisting of the combinations of the factors substrate (carbonized rice husk and bark brown ground) and increase ash (5 mL and 10 mL of rice husk ash 5 mL and 10 mL of husk ash and brown bark no gray) with four repetitions. Each experimental unit consisted of 10 cuttings, a total of 400 cuttings. Was evaluated the characteristics percentage of rooting, number and length of roots. There was no significant interaction between the factors evaluated (substrate and ash). Among the characteristics evaluated the carbonized rice showed only differences in root length and root number, while the levels of ash showed only differences in rooting percentage. The study revealed that the dosages of ash had a positive effect on rooting of cuttings of basil, as well as the substrate prepared with rice hulls provided the best conditions for root development. Thus, the rice husk is an alternative, as well as presenting features suitable for rooting also has a low cost of acquisition, being within reach of many producers.

ABSTRACT

Estudou-se a influência do tratamento diário com filtrado aquoso de Agaricus blazei Murrill (ABM) (25mg/ml), via oral, por 17 e 57 dias, em camundongos inoculados com tumor sólido de Ehrlich (TSE) por meio da curva de crescimento tumoral, do peso relativo do tumor, da contagem de regiões organizadoras de nucléolos (AgNORs) e dos padrões histológicos das massas tumorais e linfonodos poplíteos. Os animais que ingeriram o extrato aquoso do ABM por 57 dias apresentaram menor (P<0,05) crescimento do TSE, no segundo e terceiro dias, fase inflamatória do crescimento tumoral. Quanto ao peso relativo das massas tumorais, valores de AgNORs, padrões morfoistopatológicos do TSE e ocorrência de processos metastáticos, não houve alteração significativa (P>0,05) entre os animais tratados e não tratados.


The influence of daily administration of Agaricus blazei Murrill (ABM) aqueous solution (25mg/ml) during 17 and 57 days in mice bearing solid Ehrlich tumor was studied. Tumoral growth, tumoral and spleenic relative weights, nucleoly organization regions AgNORs values and tumor and popliteal lymph nodes histopathology were daily evaluated. The animals that received ABM during 57 days showed lower values (P<0.05) of tumoral growth on second and third days after the period corresponding to the inflammatory phase of tumoral growth. The relative weight of the tumor and AgNORs values were similar (P>0.05) between treated and non-treated animals. No difference in microscopic evaluation of the tumors in treated and non-treated animals was seen and metastasis in popliteal nodes of the tumor occurred in all the animals.

ABSTRACT

Cell fate decisions are governed by a complex interplay between cell-autonomous signals and stimuli from the surrounding tissue. In vivo cells are connected to their neighbors and to the extracellular matrix forming a complex three-dimensional (3-D) microenvironment that is not reproduced in conventional in vitro systems. A large body of evidence indicates that mechanical tension applied to the cytoskeleton controls cell proliferation, differentiation and migration, suggesting that 3-D in vitro culture systems that mimic the in vivo situation would reveal biological subtleties. In hematopoietic tissues, the microenvironment plays a crucial role in stem and progenitor cell survival, differentiation, proliferation, and migration. In adults, hematopoiesis takes place inside the bone marrow cavity where hematopoietic cells are intimately associated with a specialized three 3-D scaffold of stromal cell surfaces and extracellular matrix that comprise specific niches. The relationship between hematopoietic cells and their niches is highly dynamic. Under steady-state conditions, hematopoietic cells migrate within the marrow cavity and circulate in the bloodstream. The mechanisms underlying hematopoietic stem/progenitor cell homing and mobilization have been studied in animal models, since conventional two-dimensional (2-D) bone marrow cell cultures do not reproduce the complex 3-D environment. In this review, we will highlight some of the mechanisms controlling hematopoietic cell migration and 3-D culture systems.

ABSTRACT

Um panorama do consumo de fitoterápicos em farmácias comunitárias de Belo Horizonte (MG) é utilizado neste artigo como instrumento para analisar a influência da legislação nacional sobre este mercado. Para tal, foram entrevistados indivíduos que adquiriram fitoterápicos nos estabelecimentos farmacêuticos visitados. Observa-se que, na mesma nação detentora da maior biodiversidade do planeta e privilegiada por uma cultura popular conhecedora das propriedades terapêuticas das plantas, existe uma tendência de mercado para a utilização de plantas exóticas, oriundas de países desenvolvidos. A influência da legislação brasileira de fitoterápicos, que prima pela qualidade, eficácia e segurança desses medicamentos, mas que, ao mesmo tempo, tende a proscrever as drogas nativas que fazem parte da medicina popular brasileira, é discutida nesse trabalho. Verifica-se, assim, a pertinência de se discutir o perfil de usuário de fitoterápicos à luz das normas de registro desses fármacos no país no campo da saúde pública, tendo em vista que essa legislação repercute nas estratégias e/ou empecilhos para aumentar o acesso a medicamentos pela população.


An overview of the herbal drugs consumption in communitarian pharmacies in Belo Horizonte (MG), Brazil is used in this article as a tool to analyze the influence of the national law over this market. In order to reach this goal, individuals that had bought herbal drugs were interviewed at the pharmacies. It was observed that, in the same nation that has the highest biodiversity in the planet and is privileged by a popular culture knowledgeable in the therapeutic properties of the plants, there is a tendency in the market for the use of exotic plants, coming from developed countries. The influence of the Brazilian law about the herbal drugs, which constantly worries about the quality, efficiency and safety of these medicines, but at the same time, tends to proscribe the native drugs that are part of the popular medicine, is discussed in this assignment. It is concluded that it is pertinent to discuss the profile of the herbal drugs users in the spot of the laws of the country related to these drugs, in the field of the public health, since this legislation is reflected in the strategies and/or barriers to increase the access to the drugs by the population.

Este trabalho teve como objetivo a utilização dos extratos aquosos das plantas Curcuma longa e Kalanchoe brasiliensis na terapêutica tópica complementar do envenenamento botrópico experimental em camundongos, visando a antagonização dos efeitos locais (edema, hemorragia e necrose) provocados pelo veneno. O experimento mostrou que os melhores resultados foram obtidos com o extrato de Kalanchoe brasiliensis.


In this contribution, an aqueous extract from rhizomes of Curcuma longa and an aqueous extract from the aerial parts of Kalanchoe brasiliensis were investigated for their properties against the local effects (edema, hemorrhage and necrosis) of Bothrops alternatus venom in mice. The experiment showed that the best results were obtained with Kalanchoe brasiliensis extract.

Este trabalho teve como objetivo avaliar a utilização do Agaricus blazei nas formas de filtrado e suspensão total aquosa (10mg/animal) na terapêutica de camundongos portadores de tumor de Ehrlich sólido, testando sua atividade anti-neoplásica. Os animais tratados diariamente com A. blazei apresentaram valores maiores dos parâmetros hematológicos (eritrograma e leucograma), e peso relativo final do baço quando comparados com o grupo controle (água destilada), porém sem diferença significativa (P>0,05).


The aim of this study was to evaluate the antineoplastic properties of the aqueous suspension and filtrate (10mg/animal), Agaricus blazei on the solid Ehrlich tumor in mice. Groups treated with A. blazei had higher values of eritrogram and leukogram, and higher relative final spleen weight when compared to control (distillated water), but not statistically different (p>0,05).

Este trabalho teve como objetivo avaliar a utilização do cogumelo Agaricus blazei Murril (5 por cento) na terapêutica tópica do envenenamento experimental de coelhos por Bothrops alternatus, visando a antagonização dos efeitos locais (edema, hemorragia e necrose) provocados por esse veneno. Os resultados finais mostraram um menor grau de edema e do halo hemorrágico no grupo tratado quando comparados com o grupo controle (salina).


In this contribution, the mushroom Agaricus blazei Murril was investigated for its properties against the local effects of Bothrops alternatus venom in rabbits to antagonize the local effects (edema, hemorrhage and necrosis). The results showed that the group treated with Agaricus blazei had smaller edema and hemorrhage than Control group (saline).

ABSTRACT

We developed an efficient method to prepare a hybrid inorganic-organic composite based on polyvinyl alcohol (PVA) and polysiloxane using the sol-gel disc technique. Antigen obtained from Yersinia pestis was covalently immobilized onto these discs with glutaraldehyde and used as solid phase in ELISA for antibody detection in serum of rabbits experimentally immunized with plague. Using 1.25 æg antigen per disc, a peroxidase conjugate dilution of 1:4,000 and a serum dilution of 1:200 were adequate for the establishment of the procedure. These values are similar to those used for PVA-glutaraldehyde discs, plasticized filter paper discs and the polyaniline-Dacron composite discs. This procedure is comparable to that which utilizes the adsorption of the antigen to conventional PVC plates, with the amount of antigen being one fourth that employed in conventional PVC plates (5 æg/well). In addition to the performance of the polysiloxane/PVA-glutaraldehyde disc as a matrix for immunodetection, its easy synthesis and low cost are additional advantages for commercial application

ABSTRACT

This work proposes the application of the thermogravimetry (TG) in the control of quality of Cissampelos sympodialis, known popularly as 'milona'. After the extraction process, the alkaloids became the majority substances and the extracts showed the kinetic behavior of the bisbenzylisoquinoline alkaloids. This seems to be an important and useful parameter for the quality control of the 'milona' extracts.

ABSTRACT

Eggplant (Solanum melongena) is consumed extensively in Brazil. It has been believed that infusion of a powdered preparation of the fruit may reduce serum cholesterol. However, there are few documented reports on its effects on cholesterol metabolism and its possible hypocholesterolemic effect has not been proved by well-controlled studies. The aim of the present study was to observe the effects of S. melongena on the serum cholesterol and triglycerides of 38 hypercholesterolemic human volunteers ingesting S. melongena infusion for five weeks. Thirty-eight hypercholesterolemic subjects receiving either S. melongena infusion (N = 19) or placebo (N = 19) participated in two clinical experiments in which the effect of S. melongena infusion was studied with (N = 16) or without (N = 38) dietary orientation. Total cholesterol and its fractions, triglycerides, and apolipoproteins A and B were measured in blood at the beginning of the experiment and three and five weeks thereafter. No differences were observed compared to control. Intraindividual analysis showed that S. melongena infusion significantly reduced the blood levels of total and LDL cholesterol and of apolipoprotein B. After dietary orientation, no intra- or intergroup differences were seen for any of the parameters analyzed. The results suggest that S. melongena infusion had a modest and transitory effect, which was not different from that obtained with standard orientation for dyslipidemia patients (diet and physi

ABSTRACT

Due to the inadequate use of stethoscopic eartips and the possibility of they being a source of infection of the ear, a microbiologic study has been carried out on 34 stethoscopes. The material from the eartips was cultured, one eartip being "contaminated", and the other disinfected previously with 70 per cent aqueous alcohol. Microorganisms developed in 100 per cent of the contaminated eartips and in 64.71 per cent of the clean ones. Howver ther was no statistically significant difference between the two groups, showing that 70 per cent aqueous alcohol is not an efficient disinfectant

ABSTRACT

Crithidia fasciculata is an important trypanosomatid parasite commonly affecting insects and is used extensively as a model for the study of the biochemistry, ultrastructure and organization of the kDNA network of trypanosomatids. The present study describes the evolution of UV-induced morphological changes detectable by transmission electron microscopy in Crithidia fasciculata. Although only rare and minor changes in Kinetoplast DNA were demonstrable 7 h after UV irradiation, alterations of this orgtanelle were present in almost al flagellates observed 24 h and 48 h after irradiation. Other cell structures were apparently undamaged. Ultrastructural changes in kDNA did not correspond to changes in antigenicity of protein bands in western blotting against serum from Chagas' disease patients or in the presence of 3 different lectin receptors on the surface of the parasite

ABSTRACT

The effects of short ultraviolet (253 mm) radiation on the growth of the trypanosomatid flagellate Crithidia fasciculada were studied in liquid medium and nutrient agar plates. Cell duplication was completely inhibited after exposure of the flagellates to doses equal to or higher than 50 J/m2. The UV-induced lag period was dose-dependent. Survival was reduced to 1% after exposure of the parasites to 200 J/m2. Ultrastructural changes after the lag period were studied by transmission electron microscopy. Changes of the kinetoplast network structure and sometimes of the mitochondrial matrix were observed. The existence of DNA repair mechanisms in this protozoan is discussed

Os adenomas prolactinicos da hipofise sao responsaveis por grande numero de casos de amenorreia e galactorreia com ou sem alteracoes da sela turcica e por alguns casos de impotencia sexual no homem.O exame neurologico e a avaliacao endocrinologica sao de grande importancia para a conduta terapeutica. Muitos casos sao vantajosamente tratados cirurgicamente utilizando-se tecnica microcirurgica com auxilio do microscopio cirurgico e a via de acesso transesfenoidal. Os resultados sao bastante satisfatorios e a mortalidade gira em torno de 1%
